Can we over coat latex paint on a floor to help protect it?

You can but it doesn't make sense. Use a better grade of paint or put more coats. Also is you use an oilbased over coat, like a polyurethane, it will amber over time and change the original color. You are better off using a urethane based floor coating colored to your desired color.
